The closure restrictions are expected to go into effect in the Cardiff and Swansea county areas and also in the city of Llanelli.
It will take effect in Llanelli, Carmarthenshire on Saturday at 18:00 BST and in the two largest cities in Wales on Sunday from 18:00.
It will mean that eight counties will be blocked and one city in the ninth.
On Thursday, there were 25 new cases in Carmarthenshire, 39 in Cardiff and 41 in Swansea.
People in Cardiff and Swansea should not “treat this weekend as a final blast,” said Welsh Health Minister Vaughan Gething.
He also said the cases in Carmarthenshire had been “overwhelmingly” linked to Llanelli, with eight out of 10 cases there.
The Llanelli enclosure is the first to fail to cover an entire area of local authority. A zip code finder will be available online to clarify which areas are blocked, Gething said.
Most of the cases in Llanelli have been linked to people who socialize without social distancing.
Burry Port will not be included in the restricted area.
People living in Caerphilly, Rhondda Cynon Taf, Bridgend, Blaenau Gwent, Merthyr Tydfil, and Newport are no longer allowed to leave their areas without a reasonable excuse, under current rules.
The changes mean that around 1.5 million people will be under local lockdown, just under half of the Welsh population.
