Hydroxychloroquine and azithromycin, which have been proposed for the treatment of COVID-19 patients, are potentially lethal combination, and can have a serious impact on the cardiovascular system, according to a study.
The researchers, including from Vanderbilt University and Stanford University in the united states, carried out an observational, retrospective study meta-analysis of a data base covering more than 21 million of adverse event case reports.
The reports cover all classes of medications in more than 130 countries, including the November 14, 1967, and March 1, 2020 and, above all, before the COVID-19 pandemic.
The study, published in the journal Circulation, compared the cardiovascular adverse drug reactions (CV-Adrs) in patients receiving hydroxychloroquine, azithromycin, or the combination of both drugs with all other drugs cardiovascular in the database.
Hydroxychloroquine and azithromycin, alone or in combination, have been proposed for the treatment of COVID-19 patients, the researchers said.
He explained that the reports of potentially lethal acute cardiac proarrhythmogenic effects-the promotion of the irregular rhythms of the heart — have been described primarily with azithromycin, but also with hydroxychloroquine.
Their combination produced a stronger signal, the scientists said. Hydroxychloroquine is also associated with potentially lethal heart failure when the exposure lasted for several months, according to the researchers.
Of the more than 21 million cases reports of adverse reactions to the medications, the researchers have extracted the reports of cases of hydroxychloroquine and azithromycin, alone or in combination.
They found that 76,822 reports of adverse events associated with hydroxychloroquine alone, and in 28.4% of cases (21,808), the drug was suspected to be associated with the adverse event.
The study found that 89,692 reports of adverse events associated with azithromycin alone, and in 60.8% of cases (54,533), the medicine was suspected to be associated with adverse events.
The researchers said that the 607 reports of adverse events associated with the combination of both drugs.
The hydroxychloroquine was significantly associated with the development of conduction disorders, and heart failure, it said.
On Monday, the World Health Organization also suspended the trial of hydroxychloroquine in COVID-19 patients due to safety concerns, the Director General Tedros Adhanom Ghebreyesus, said.
